目的:观察药对荆芥-防风的挥发油、含油水煎液、去油水煎液对内毒素(LPS)诱导的急性肺损伤(ALI)小鼠模型的影响,进一步揭示该药对的抗炎作用及物质基础。方法:采用小鼠ip内毒素(LPS,5 mg·kg-1)制备ALI模型。除地塞米松组(5 mg·kg-1)ip给药1次外,其余各组小鼠均连续ig给药5 d,每日1次,末次给药30 min后造模(空白组不进行造模)。注射LPS 6 h后,小鼠取血分离血清,并收集支气管肺泡灌洗液(BALF),剖取肺组织。测定小鼠全血白细胞分类计数,小鼠肺指数、肺湿/干重(W/D),BALF中总蛋白含量,肺组织髓过氧化物酶(MPO)活性、丙二醛(MDA)含量;血清白细胞介素6(IL-6)、白细胞介素10(IL-10)、肿瘤坏死因子α(TNF-α)水平。结果:药对荆芥-防风挥发油(45.19μL·kg-1)、含油水煎液(6.67g·kg-1,含油20μL)显著降低ALI小鼠全血白细胞(WBC)、淋巴细胞(LY)计数,降低肺指数、肺W/D,降低肺组织MPO活性与MDA含量及血清IL-10,TNF-α含量;去油水煎液(6.67 g·kg-1)明显降低全血WBC,LY计数,肺W/D及肺组织MDA含量。挥发油对血清IL-6水平亦显著降低。结论:药对荆芥-防风的挥发油、含油水煎液对ALI小鼠肺组织的炎性病变有较好的对抗作用,表现出抗炎效应,作用机制与抑制炎性细胞因子释放、抗氧化有关,其中药对挥发油部位可认为是其发挥抗炎效应的重要物质基础之一。
