目的为了解大肠癌p53基因突变与p53基因蛋白表达之间的关系。方法用RT-PCR-SSCP方法检测100例新鲜大肠腺癌组织P53基因突变,以PAb1801单克隆抗体(单抗)免疫组织化学(LSAB法)染色检测相同腺癌组织p53基因蛋白表达。结果在这100例大肠腺癌中,p53基因突变阳性率为51%,PAb1801单抗免疫组织化学染色(免疫级化)阳性率为62%。在p53基因突变检测阴性的49例中,24例PAb1801阴性,25例PAb1801阳性;51例p53基因突变检测阳性中,14例PAb1801阴性,37例PAb1801阳性,Fisher'sexacttestP=0.02。结论大肠腺癌p53基因突变是参与和影响p53基因蛋白表达的一个主要因素。
Objective To understand the relationship between p53 gene mutation and p53 gene protein expression in colorectal cancer. Methods RT-PCR-SSCP method was used to detect the mutations of P53 gene in 100 cases of fresh colorectal adenocarcinoma and the expression of p53 gene protein in the same adenocarcinoma was detected by monoclonal antibody (monoclonal antibody) immunohistochemistry (LSAB) staining with PAb1801. Results In these 100 cases of colorectal adenocarcinoma, the positive rate of p53 gene mutation was 51%, and the positive rate of PAb1801 monoclonal antibody immunohistochemistry (immunological grade) was 62%. Of the 49 patients with negative p53 mutations, 24 were negative for PAb1801 and 25 were positive for PAb1801. Of the 51 patients with positive p53 mutations, 14 were negative for PAb1801, 37 were PAb1801 positive, and Fisher’s exact test P=0.02. Conclusion p53 gene mutation in colorectal adenocarcinoma is a major factor involved in the expression of p53 gene protein.
