用常规方法免疫得到的抗人心肌肌球蛋白轻链(CM-LC)的抗血清,常与骨骼肌及平滑肌肌球蛋白轻链有较强的交叉反应。单克隆抗体因系特异地识别CM-LC上的单一抗原决定簇,因而能克服这一困难。我们应用淋巴细胞杂交瘤技术已经得到四株分泌抗人CM-LC单克隆抗体的杂交瘤细胞株,并对其中三株(1G_6、2B_4、2F_6)进行了克隆。它们与人骨骼肌及平滑肌肌球蛋自轻链的交叉反应分别为2F_6 28.6%及15.6%;2B_4 45%及81%;1G_6对二者的交叉反应均为81%。2F_6属小鼠IgG亚类IgG_3;2B_4及1G_6则均为IgG_1。
The anti-human myocardial myosin light chain (CM-LC) antiserum obtained by conventional immunization often has a strong cross-reaction with skeletal and smooth muscle myosin light chains. Monoclonal antibodies overcome this difficulty by specifically recognizing a single antigenic determinant on the CM-LC. We have obtained four hybridoma cell lines secreting anti-human CM-LC monoclonal antibodies using lymphocyte hybridoma technology and cloned three of them (1G_6,2B_4,2F_6). Their cross-reactivity with human skeletal muscle and smooth muscle myoglobin from the light chain were 2F_6 28.6% and 15.6%, 2B_4 45% and 81%, respectively. The cross-reactivity of 1G_6 to both of them was 81%. 2F_6 is a mouse IgG subclass IgG_3; 2B_4 and 1G_6 are IgG_1.
