目的:观察小脑顶核刺激(FNS)对急性心肌梗死(AMI)大鼠自主神经功能的影响。方法:成年雄性Sprague-Dawley(SD)大鼠100只,分为:①AMI组(30只),单纯结扎大鼠左冠状动脉(LCA);②FNS组(30只):结扎大鼠LCA,1h后电刺激左侧小脑顶核1h;③FNL组(30只),鹅膏氨酸毁损两侧小脑顶核,5d后再按②方法操作;④Sham组(10只),即假手术组,开胸手术后仅在LCA下穿线但不结扎,电极插入小脑顶核,不予刺激。各组在开胸术后6h、24h、7d、21d4个时间点,进行心率变异性(HRV)的频域分析。21d记录心脏血流动力学参数并计算心肌梗死面积。结果:结扎6h后FNS组频域分析指标-低频(LowFrequency,LF)低于AMI和FNL组(P<0.01),同时FNS组高频(HighFrequency,HF)明显高于其他干预组(P<0.01)。结扎1d后,FNS组仍然呈现HF成分的增加和LF成分的下降(P<0.05)。7d后只有假手术组保留了较高的HF和较低的LF,各干预组之间的差别消失。21d后各组之间HRV参数无差别。FNL组与AMI组比较,各时间点HRV参数差异无统计学意义(P>0.05)。结扎后21d比较各组血流动力学指标,心肌梗死面积也无差别(P>0.05)。结论:FNS可改善AMI后大鼠心脏神经功能。
Objective: To observe the effects of fastigial nucleus stimulation (FNS) on autonomic nervous system in rats with acute myocardial infarction (AMI). METHODS: One hundred male Sprague-Dawley rats were divided into three groups: ①AMI group (n = 30), left coronary artery (LCA) group; ②FNS group (n = 30) The left parietal fastigial nucleus was stimulated by electrical stimulation for 1 h. ③FNL group (n = 30) and euthymic acid lesioned the nucleus parietalis on both sides, and then operated by ② method after 5 days. ④Sham group (10), sham operation group, Only after LCA threading but not ligation, the electrode inserted into the cerebellar fastigial nucleus, without stimulation. The frequency domain analysis of heart rate variability (HRV) was performed at 6h, 24h, 7d and 21d after thoracotomy in each group. 21d Cardiac hemodynamic parameters were recorded and myocardial infarct size was calculated. Results: After 6 hours of ligation, the frequency of low-frequency (LF) analysis of FNS group was lower than that of AMI and FNL groups (P <0.01), while the frequency of high frequency of FNS group was significantly higher than that of other intervention groups (P <0.01) ). After 1 day of ligation, the FNS group still showed the increase of HF and the decrease of LF (P <0.05). Only the sham-operated group retained higher HF and lower LF after 7d, and the differences between the intervention groups disappeared. There was no difference in HRV parameters between groups after 21 days. There was no significant difference in HRV parameters between FNL group and AMI group at each time point (P> 0.05). After 21 days of ligation, hemodynamic parameters were compared between groups, and infarct size was also not different (P> 0.05). Conclusion: FNS can improve cardiac function after AMI in rats.
