分别采用平衡法和摇瓶法测定高、中、低纯度(71.0%、49.2%和25.4%)吴茱萸提取物中吴茱萸碱和吴茱萸次碱的溶解度及表观分配系数,采用HPLC法测定两者的浓度。高中低纯度吴茱提取物中吴茱萸碱和吴茱萸次碱均在PEG-400中溶解度最高,分别为8~14 mg/ml和13~19 mg/ml;异丙醇中的溶解度最低(均小于7 mg/ml)。吴茱萸碱和吴茱萸次碱在介质中的溶解度依次为PEG-400>Tween-80>RH-40>MCT>无水乙醇≈1,2-丙二醇≈1,3-丁二醇≈异丙醇。两者的表观分配系数的对数值(lgP)为0.6~1.2。吴茱萸提取物纯度越高,吴茱萸碱和吴茱萸次碱的溶解度越低;纯度与表观分配系数无相关性。pH对溶解度和表观分配系数均无明显影响。
The solubility and apparent partition coefficient of evodiamine and rutaecarpine in Evodia rutaecarpa extract with high, medium and low purity (71.0%, 49.2% and 25.4%) were determined by equilibrium method and shake flask method, respectively, concentration. Evodiamine and rutaecarpine were the highest in PEG-400, which were 8-14 mg / ml and 13-19 mg / ml, respectively. The solubility of isopropanol was the lowest (all less than 7 mg / ml). The evusate of evodiamine and rutaecarpine in the medium were PEG-400> Tween-80> RH-40> MCT> anhydrous ethanol ≈1,2-propanediol ≈1,3-butanediol ≈ isopropanol. The logarithm of the apparent partition coefficient (lgP) of both is 0.6-1.2. The higher the purity of Evodia rutaecarpa extract, the lower the solubility of evodiamine and rutaecarpine; the purity has no correlation with the apparent partition coefficient. No significant effect of pH on solubility and apparent partition coefficient.
