目的 :探讨应用参考链构象分析法对HLA DRB1等位基因分型的准确性及实用性。方法 :应用毛细管参考链介导的构象分析法建立 31个等位基因的标准迁移率 ,对 5 0例样本进行分型 ,并与SBT法检测结果进行比较。结果 :5 0例样本中 4 7 5 0与测序法测得的等位基因一致 ,以DRB1 0 80 32、DRB1 0 90 12为参考链无法检测出DRB1 110 1、DRB1 14 0 3。结论 :毛细管参考链介导的构象分析法具有准确性高、分辨率高、成本低、高通量的特点 ,值得在HLA分型中应用推广
Objective: To investigate the accuracy and practicability of HLA DRB1 allele typing by using conformational analysis of reference chain. Methods: The standard mobility of 31 alleles was established by conformational analysis mediated by capillary reference chain. Fifty samples were typed and compared with the SBT test results. Results: The 4 7 5 0 of 50 samples was consistent with the allele measured by sequencing. DRB1 110 1 and DRB1 14 0 3 could not be detected by DRB1 0 80 32 and DRB1 0 90 12. Conclusion: The conformational analysis method mediated by capillary reference chain has the characteristics of high accuracy, high resolution, low cost and high throughput. It is worth to popularize in HLA typing
