目的探讨整合素A2(ITGA2)调控卵巢癌细胞系锌指增强子结合蛋白1(ZEB1)的表达对卵巢癌转移侵袭能力的影响。方法分析490例卵巢癌标本中ITGA2与ZEB1 mRNA表达的相关性;应用Western blot比较4种卵巢癌细胞系中ITGA2与ZEB1蛋白水平的相关性;将针对ZEB1的siRNA转染到SKOV3细胞系中,Western blot验证ZEB1蛋白表达变化,Transwell验证其转移侵袭能力的变化;将针对ITGA2的siRNA转染到SKOV3细胞系,Western blot验证其ITGA2及ZEB1的蛋白表达变化,Transwell验证其转移侵袭能力的变化。结果 490例卵巢癌标本中,ZEB1与ITGA2在mRNA水平呈显著正相关(P<0.01);卵巢癌细胞系中ZEB1和ITGA2蛋白水平呈正相关;下调ZEB1后,卵巢癌细胞侵袭转移能力降低;下调ITGA2后,ZEB1表达降低,卵巢癌细胞转移侵袭能力也降低。结论 ITGA2正调控ZEB1的表达,促进卵巢癌细胞转移侵袭,提示降低ITGA2和ZEB1的表达可抑制卵巢癌细胞的转移侵袭。
Objective To investigate the effect of integrin A2 (ITGA2) regulating the expression of zinc finger enhancer binding protein 1 (ZEB1) in ovarian cancer cells on the invasion and metastasis of ovarian cancer. Methods The correlation between ITGA2 and ZEB1 mRNA expression in 490 cases of ovarian cancer was analyzed. The correlation between ITGA2 and ZEB1 protein levels in four ovarian cancer cell lines was analyzed by Western blot. The siRNA targeting ZEB1 was transfected into SKOV3 cell line, The changes of ZEB1 protein expression were detected by Western blot, and the changes of invasion and metastasis were verified by Transwell. The siRNA targeting ITGA2 was transfected into SKOV3 cell line. The protein expression of ITGA2 and ZEB1 was verified by Western blot. Results In 490 specimens of ovarian cancer, there was a significant positive correlation between ZEB1 and ITGA2 at the mRNA level (P <0.01). There was a positive correlation between ZEB1 and ITGA2 protein levels in ovarian cancer cells. The down-regulation of ZEB1 could reduce the invasion and metastasis of ovarian cancer cells. After ITGA2, ZEB1 expression decreased, ovarian cancer cell invasion and metastasis also decreased. Conclusion ITGA2 positively regulates the expression of ZEB1 and promotes the invasion and metastasis of ovarian cancer cells, suggesting that reducing the expression of ITGA2 and ZEB1 can inhibit the metastasis and invasion of ovarian cancer cells.
