目的:研究人脑老化过程中胶质纤维酸性蛋白(GFAP)的表达变化。方法:两例人脑标本,样本一:女性,64岁,生前诊断有血管性淀粉病变;样本二:男性,16岁,死于急性心肌炎,两例标本均于死后12 h取脑,取皮质和海马部位,进行GFAP的常规免疫组化染色进行定量分析。结果:老年脑星形胶质细胞呈明显激活状态,GFAP阳性细胞数目显著增多,胞体增大,突起粗短,刚果红染色可见老年斑周围GFAP阳性细胞成簇聚集;年轻脑GFAP阳性细胞数目少,胞体瘦小,突起细长,未见GFAP阳性细胞成簇聚集。统计学分析表明GFAP阳性细胞数和蛋白质的表达变化在老年脑和年轻脑具有差别显著性(P<0.01)。结论:衰老和血管淀粉样病变在人脑星形胶质细胞的增生和激活过程中具有重要的作用。
Objective: To study the expression of glial fibrillary acidic protein (GFAP) in human brain during aging. Methods: Two human brain specimens, sample 1: female, aged 64, were diagnosed with vascular amyloidosis during their lifetime. Sample 2: male, aged 16, died of acute myocarditis. Both samples were taken at 12 h after death Cortical and hippocampal parts of GFAP routine immunohistochemical staining for quantitative analysis. Results: The number of GFAP positive cells increased significantly, the number of GFAP positive cells increased, the number of GFAP positive cells increased and the length of protuberances increased. The number of GFAP positive cells around the senile plaques was clustered and aggregated by congo red staining. Small body cells, protruding slender, no GFAP positive cells clustered. Statistical analysis showed that the number of GFAP positive cells and protein expression changes in the elderly brain and young brain was significantly different (P <0.01). Conclusion: Aging and vascular amyloid lesions play an important role in the proliferation and activation of human astrocytes.
