目的:明确红曲对去卵巢大鼠骨质疏松模型血清ALP,TRAP及骨组织TNF-α,RANK表达的影响。方法:取12周龄Wistar雌性大鼠40只,随机分为结合雌激素组、红曲组、模型组、假手术组,每组10只。前三组行完整双侧卵巢摘除术,假手术组仅打开腹腔,不切除卵巢。灌胃12周后,处死各组动物,采用多光谱成像系统检测骨密度,用ELISA法测定各组大鼠血清中ALP及TRAP,用Image Pro Plus对免疫组化染色图像结果进行定量分析。结果:红曲组TNF-α免疫组化表达水平低于模型组(P<0.05),红曲组RANK免疫组化表达水平低于模型组(P<0.05)。结论:红曲可以提高血清ALP表达水平,降低血清TRAP表达水平,能够通过降低TNF-α水平及RANK水平来提高去卵巢大鼠骨密度,其机理与雌激素类似,可能与成骨细胞及破骨细胞活跃程度有关。
Objective: To determine the effects of Monascus on serum ALP, TRAP and the expression of TNF-α and RANK in osteoporosis model of ovariectomized rats. Methods: Forty Wistar female rats of 12 weeks old were randomly divided into estrogen group, Hong song group, model group and sham operation group, with 10 rats in each group. The first three groups of complete bilateral ovariectomy, sham-operated group only open the abdominal cavity, do not remove the ovary. After 12 weeks of gavage, each group of animals was sacrificed, and the bone mineral density (BMD) was measured by multispectral imaging system. The serum ALP and TRAP were detected by ELISA. The results of immunohistochemistry were analyzed quantitatively by Image Pro Plus. Results: The expression level of TNF-α in Hongqu songts group was lower than that in model group (P <0.05). The expression of RANK in Hongqu songts group was lower than that in model group (P <0.05). CONCLUSION: Monascus can increase the expression of serum ALP and decrease the expression of TRAP in serum. It can increase the bone density of ovariectomized rats by decreasing the levels of TNF-α and RANK. The mechanism may be similar to that of estrogen, The degree of bone cell activity.
