以多糖得率为指标,选用正交试验对紫茎泽兰粗多糖的提取工艺进行优化,再经乙醇沉淀、脱蛋白和真空干燥后得到紫茎泽兰多糖(EAP)。结果表明紫茎泽兰粗多糖最佳提取工艺为:温度为100℃,在料液比1:20下提取2 h,提取2次。用脱氧核糖体系测EAP对·OH的清除作用;用硫代巴比妥酸法测小鼠肝组织丙二醛;用分光光度法测EAP对邻苯三酚自氧化体系的抑制作用。结果表明:EAP具有抗氧化活性,以抑制体外脂质过氧化效果最为显著。
Using polysaccharide yield as an index, orthogonal test was used to optimize the extraction process of crude polysaccharide from Eupatorium adenophorum. Eupatorium adenophorum polysaccharide (EAP) was obtained after ethanol precipitation, deproteinization and vacuum drying. The results showed that the optimal extraction process of crude polysaccharide from Eupatorium adenophorum was as follows: extraction temperature was 100 ℃, extracting for 2 h at the ratio of material to liquid 1:20 and extracting twice. The scavenging effect of EAP on · OH was determined by deoxyribose system. The malondialdehyde (MDA) in mouse liver tissue was measured by thiobarbituric acid method. The inhibitory effect of EAP on autoxidation of pyrogallol was determined by spectrophotometry. The results showed that: EAP has antioxidant activity, in order to inhibit lipid peroxidation in vitro the most significant.
