目的 :观察钩端螺旋体 DNA疫苗的保护效果 ,并探讨该疫苗的免疫机制。方法 :应用定向克隆的方法构建 DNA疫苗 pc D- fla B,肌肉途径免疫豚鼠 ,观察豚鼠攻击感染钩体后的保护率 ,以 EL ISA法检测特异性抗体 Ig G水平 ,并观察疫苗对豚鼠腹腔巨噬细胞分泌的 TNF活性的影响。结果 :该疫苗对同型钩体的保护率为 10 0 % ,特异性抗体水平在疫苗注射第 6周达到高峰 ,TNF的活性明显增高。 结论 :DNA疫苗 pc D- fla B对同型钩体感染有较强的保护作用 ;能够有效地诱导机体的体液免疫应答 ,并增强巨噬细胞所产生的 TNF的活性。
Objective: To observe the protective effect of Leptospira DNA vaccine and to explore the immune mechanism of the vaccine. Methods: The DNA vaccine pc D-fla B was constructed by directional cloning method. The guinea pigs were immunized by muscle route. The protection rate of guinea pigs after challenge with infected leptospira was observed. The level of Ig G was detected by ELISA. Effect of macrophage secretion of TNF activity. Results: The protective rate of the vaccine against the same type of leptospira was 100%. The level of the specific antibody peaked at the sixth week of the vaccine injection, and the activity of TNF was significantly increased. CONCLUSION: DNA vaccine pc D-fla B has a strong protective effect against leptospira infection; it can effectively induce the humoral immune response and enhance the activity of TNF produced by macrophages.
