目的:建立能同时测定大鼠血浆中马钱子碱和士的宁浓度的HPLC分析方法。方法:大鼠血浆样品以石杉碱甲为内标,液液萃取法处理后用Kromasil C18柱分离,以乙腈-0.01 mol.L-1庚烷磺酸钠与0.02 mol.L-1磷酸二氢钾等量混合后用10%磷酸调体系pH 2.8(24∶76)作流动相,检测波长264 nm。结果:血浆中杂质不干扰样品的含量测定,马钱子碱和士的宁在50~2 000μg.L-1线性良好,平均提取回收率均大于86%,日内、日间精密度均小于8%,稳定性符合体内药物分析要求。结论:方法灵敏度高、精密度好,可应用于马钱子总生物碱中主要成分的药物动力学研究。
OBJECTIVE: To establish an HPLC method for the simultaneous determination of strychnine and strychnine in rat plasma. METHODS: Rat plasma samples were treated with huperzine A as an internal standard and were separated by a Kromasil C18 column using a liquid-liquid extraction method. The samples were eluted with acetonitrile-0.01 mol. L-1 heptane sulfonate and 0.02 mol·L-1 di Hydrogen and potassium mixed with 10% phosphoric acid system pH 2.8 (24:76) as the mobile phase, detection wavelength 264 nm. Results: The contents of impurities in the plasma did not interfere with the determination of the samples. The linearities of strychnine and strychnine at 50 ~ 2 000μg.L-1 were good with the average recoveries of more than 86%. The intra- and inter-day precision were less than 8 %, Stability in line with in vivo drug analysis requirements. Conclusion: The method has high sensitivity and good precision and can be applied to the pharmacokinetic study of the main components of total alkaloids in Strychnos nux - vomica.
