目的 :研究雷公藤多甙和 IL - 10对人树突状细胞 (DC)表面人类白细胞抗原 - DR(HL A- DR)和 CD80表达及 IL - 12 p40转录和分泌的影响。方法 :通过粒细胞 -巨噬细胞型集落刺激因子 (GM- CSF)、IL- 4和肿瘤坏死因子 α(TNF- α)体外培养体系 ,从人外周血单个核细胞中获得 DC,细胞表面 HL A- DR和 CD80表达采用流式细胞仪分析 ,IL - 12 p40转录和分泌分别采用逆转录多聚酶链反应 (RT- PCR)技术和 EL ISA法。结果 :雷公藤多甙 (5~ 2 0μg/ ml)、IL - 10 (5 0~ 2 0 0 ng/ ml)能下调 DC表面 HL A- DR和 CD80的表达 ,同时雷公藤多甙、IL- 10能抑制 DC内 IL- 12 p40 m RNA的转录和分泌。结论 :雷公藤多甙、IL- 10能通过抑制表面分子的表达和细胞因子的合成而干扰 DC的成熟和功能。
Objective: To study the effects of Tripterygium glycosides and IL - 10 on the expression of HLA - DR and CD80 and the transcription and secretion of IL - 12 on human dendritic cells. Methods: DCs were obtained from human peripheral blood mononuclear cells by in vitro culture system of granulocyte-macrophage colony-stimulating factor (GM-CSF), IL-4 and tumor necrosis factor-α (TNF- The expression of A-DR and CD80 were analyzed by flow cytometry. The transcription and secretion of IL-12 p40 were detected by RT-PCR and ELISA, respectively. Results: Tripterygium glycosides (5 ~ 20 μg / ml) and IL - 10 (50 ~ 200 ng / ml) could downregulate HL-DR and CD80 expression on DCs. 10 inhibits the transcription and secretion of IL-12 p40 m RNA in DCs. Conclusion: Tripterygium glycosides and IL-10 can interfere with the maturation and function of DC by inhibiting the expression of surface molecules and the synthesis of cytokines.
