利用RT-PCR扩增12例B-CLL外周血单个核细胞的TCR Vβ基因24个亚家族的互补决定区3(CDR3),以了解T细胞的表达情况。PCR产物进一步经基因扫描分析CDR3长度而确定T细胞的克隆性。结果显示:12例B-CLL共表达13个TCRVβ亚家族T细胞,每例病人仅表达3-9个Vβ亚家族,主要为Vβ2,Vβ3,Vβ6,Vβ7,Vβ17或Vβ19,9例病人的一个或两个Vβ3家族T细胞里克隆性生长。提示B-CLL选择性地表达TCR VβT细胞,大部分B-CLL出现克隆性增殖T细胞,这可能是对白血病细胞相关抗原的免疫反应。
RT-PCR was used to amplify the complementarity determining region 3 (CDR3) of 24 subfamilies of TCR Vβ gene in peripheral blood mononuclear cells of 12 patients with B-CLL to understand the expression of T cells. The PCR product was further subjected to a gene scan to analyze the length of the CDR3 to determine the clonality of the T cells. The results showed that 12 cases of B-CLL co-expressed 13 TCR Vβ subfamily T cells, and each patient only expressed 3-9 Vβ subfamily, mainly Vβ2, Vβ3, Vβ6, Vβ7, Vβ17 or Vβ19, one of 9 patients. Or clonal growth in two Vβ3 family T cells. It is suggested that B-CLL selectively expresses TCR Vβ T cells, and most of B-CLL appear clonally proliferating T cells, which may be an immune response to leukemic cell-associated antigens.
