目的研究低钙对血清补体C3的影响及其可能的机制。方法雄性Wistar大鼠30只,按体重随机分为低钙组(喂饲0.15%Ca)和正常钙对照组(喂饲0.50%Ca),每组15只。喂养12w后,麻醉处死大鼠,取组织和血清。采用双向电泳技术分离大鼠血清,联合4700串联飞行时间质谱仪和蛋白质数据库分析鉴定差异表达蛋白质,并检测各组大鼠血清补体C3等生化指标。为通过人群进一步验证,在性别、年龄匹配的基础上,调查(1~3岁)佝偻病儿童及健康对照儿童,检测血清补体C3等生化指标。用real time PCR技术检测体外低钙培养的HepG2细胞及各组大鼠肝脏补体C3、furin酶mRNA表达情况。结果通过血清蛋白质组学技术筛选出低钙动物模型血清补体C3显著降低。与正常钙对照组(0.30±0.02)g/L比,低钙组大鼠血清补体C3(0.24±0.04)g/L显著降低(P<0.05)。共调查123名(1~3岁)佝偻病儿童和114名健康对照儿童,与健康对照儿童(1.52±0.10)g/L相比,佝偻病儿童血清补体C3(1.27±0.10)g/L显著降低(P<0.05)。低钙时HepG2细胞和大鼠肝脏补体C3、furin酶mRNA表达下降。结论低钙可能通过降低furin酶mRNA的表达,导致佝偻病儿童血清补体C3水平降低,机体免疫力下降,增加上呼吸道感染的危险。[营养学报,2012,34(6):586-590]
Objective To study the effect of low calcium on serum complement C3 and its possible mechanism. Methods Thirty male Wistar rats were randomly divided into low calcium group (fed 0.15% Ca) and normal calcium control group (fed 0.50% Ca), with 15 rats in each group. After feeding for 12 weeks, the rats were anesthetized and the tissues and serum taken. The serum of rats was separated by two-dimensional electrophoresis and the differentially expressed proteins were identified by 4700 tandem time-of-flight mass spectrometry and protein database analysis. Serum complement C3 and other biochemical markers were detected in all groups. In order to further verify the results of population screening, children with chronic rickets (aged 1 to 3 years) and healthy controls were investigated on the basis of gender and age matching, and biochemical indexes such as serum complement C3 were detected. Real-time PCR was used to detect the mRNA expression of complement C3 and furin in HepG2 cells cultured in vitro and liver in each group. Results Serum proteomics was used to screen out the serum calcium of C3 in low-calcium animal model. Compared with normal calcium control group (0.30 ± 0.02) g / L, serum complement C3 (0.24 ± 0.04) g / L in low calcium group was significantly lower (P <0.05). A total of 123 children with rickets (1 to 3 years old) and 114 healthy children were enrolled in this study. Serum complement C3 (1.27 ± 0.10) g / L was significantly lower in children with rickets than in healthy children (1.52 ± 0.10) g / L P <0.05). Low calcium calcium and HepG2 cells and rat liver complement C3, furin enzyme mRNA expression decreased. Conclusion Low calcium may reduce the level of furin mRNA expression in children with rickets, resulting in decreased serum complement C3, decreased immunity and increased risk of upper respiratory tract infection.
