目的探讨华支睾吸虫(Cs)硫氧还蛋白过氧化物酶(TPx)对脂多糖(LPS)致脓毒症小鼠的保护作用。方法 80只Balb/c雄性小鼠随机分成两组,腹腔注射LPS(4 mg/kg)制备脓毒症小鼠模型,造模后30 min尾静脉分别注射PBS、rCsTPx(2.5 mg/kg),观察注射后各组小鼠72 h死亡率;重复实验,每组分别于实验后的3、6、9、12、24、48、72 h“安乐死”处死小鼠,收集血清并取24 h组织用于病理切片观察。结果尾静脉注射CsTPx治疗的小鼠生存率由10%提高到40%;检测血清谷草转氨酶(AST)和谷丙转氨酶(ALT)水平在造模后6、9、12、24 h明显下降,并且血清中炎症因子IL-6在3、6、9 h也明显下调,差异均有统计学意义;组织HE染色揭示CsTPx治疗组小鼠肾组织损伤减轻,炎症细胞浸润也减少。结论 rCsTPx对LPS致脓毒症小鼠有较好的保护作用。
Objective To investigate the protective effect of Clonorchis sinensis (Cs) thioredoxin peroxidase (TPx) on sepsis mice induced by lipopolysaccharide (LPS). Methods Eighty Balb / c male mice were randomly divided into two groups. The mice were injected intraperitoneally with LPS (4 mg / kg), and the caudal vein was injected with PBS and rCsTPx (2.5 mg / kg) The mortality of mice in each group was observed at 72 h after injection. The mice were sacrificed at 3, 6, 9, 12, 24, 48 and 72 h after euthanasia, h tissue for pathological observation. Results The survival rate of mice treated with tail vein injection of CsTPx was increased from 10% to 40%. The levels of serum AST and ALT were significantly decreased at 6, 9, 12 and 24 h Serum inflammatory cytokines IL-6 also significantly decreased at 3,6,9 h, the differences were statistically significant; tissue HE staining revealed that renal damage in CsTPx treatment group mice reduced, infiltration of inflammatory cells also decreased. Conclusion rCsTPx has good protective effect on LPS-induced septic mice.
