目的应用PCR-DGGE技术评价西芹对昆明小鼠肠道菌群平衡的影响。方法给予小鼠高脂饲料,复制高脂模型,模型复制成功后,每日给予小鼠西芹提取液灌胃,连续28 d。分别在实验的0、14和42 d,收集每只小鼠粪便,提取基因组DNA,应用PCR-DGGE(聚合酶链式反应——变性梯度凝胶电泳)技术获得肠道菌群分子指纹图谱,针对相似性、多样性分析及优势条带的序列进行分析。结果高脂饲料喂养后肠道益生菌约氏乳杆菌等趋近消失,出现了特异菌真杆菌,而西芹灌胃组和食用基础饲料组真杆菌数量显著减少,且西芹灌胃组作用大于食用基础饲料自然恢复组。结论西芹对高脂小鼠肠道菌群的平衡有显著的恢复作用。
Objective To evaluate the effect of celery on the balance of intestinal flora in Kunming mice by PCR-DGGE. Methods The mice were given high fat diet and the model of hyperlipidemia was replicated. After the model was successfully replicated, the celery extract was given to the mice daily for 28 days. The faeces of each mouse were collected at 0, 14 and 42 days after the experiment, genomic DNA was extracted, and molecular fingerprinting of intestinal flora was obtained by PCR-DGGE (Polymerase Chain Reaction-denaturing Gradient Gel Electrophoresis) Analysis of similarities, diversity analyzes and sequences of dominant bands. Results The intestinal probiotics Lactobacillus johnsonii disappeared and the specific bacterium eubacterium was absent after high-fat diet feeding, but the number of eubacteria in celery gavage group and edible basal diet group significantly decreased, Greater than the basic diet edible natural recovery group. Conclusion Celery has a significant effect on the balance of intestinal flora in high-fat mice.
