目的:观察养肝利胆颗粒对胆固醇结石小鼠肝脏PPAR-γ及CYP7A1表达的影响。方法:38只C57BL/6雌性小鼠随机分为正常对照组(n=10)、模型组(n=15)和养肝利胆颗粒组(n=13)。其中后两组小鼠采用高脂饮食诱发法建立胆固醇结石模型。造模过程中,养肝利胆颗粒组小鼠予养肝利胆颗粒2.1g/(kg.d)灌胃治疗。8周后观察各组小鼠的成石率,并用Western Blotting法检测肝脏中PPAR-γ及CYP7A1的表达。结果:养肝利胆颗粒组成石率显著降低(P<0.01),小鼠肝脏PPARγ及CYP7A1表达增强。结论:养肝利胆颗粒可通过增强肝脏PPARγ及CYP7A1的表达,从而发挥防治胆石病的作用。
Objective: To observe the effects of Yanggan Lidan Granules on the expression of PPAR-γ and CYP7A1 in liver of cholesterol-treated mice. Methods: Thirty-eight female C57BL / 6 mice were randomly divided into normal control group (n = 10), model group (n = 15) and Yanggan Liguan granule group (n = 13). The latter two groups of mice using high-fat diet induced cholesterol gallstone model. Modeling process, the liver and gallbladder particles to the liver and gallbladder particles 2.1g / (kg.d) intragastric treatment. After 8 weeks, the rate of stone formation in each group was observed, and the expression of PPAR-γ and CYP7A1 in liver were detected by Western Blotting. Results: The stone ratio of Yanggan Liguan granule was significantly decreased (P <0.01), and the expression of PPARγ and CYP7A1 in liver increased. Conclusion: Yanggan Lidan Granule can play a role in prevention and treatment of cholelithiasis by enhancing the expression of PPARγ and CYP7A1 in liver.
