为评价CD44分子过量表达在葡萄膜与结膜黑色素瘤发生发展过程中的作用,应用流式细胞术(FCM)及细胞免疫荧光染色技术对葡萄膜恶性黑色素瘤(UMM)、结膜色素痣(CN)及恶性黑色素瘤(CMM)的CD44V表达进行定量检测。结果表明:7例CMM较5例CN的CD44V表达量明显增加(P<0.05),7例CMM及40例UMM的CD44V表达阳性率分别为71.43%及62.5%;UMM中伴巩膜受侵犯者较不伴巩膜受侵犯者的CD44V表达量明显增加(P=0.0105);UMM的CD44V表达量与增殖指数(PI)、S期细胞百分率(SPF)呈显著负相关(P<0.01;P<0.05)。以上表明CD44V过量表达与CMM及UMM的发生有关,并与UMM局部浸润有关。
To evaluate the role of overexpression of CD44 in the development of uveal and conjunctival melanomas, flow cytometry (FCM) and immunofluorescence staining were applied to uveal malignant melanoma (UMM) and conjunctival pigmentary nevus (CN). Quantification of CD44V expression in malignant melanoma (CMM). The results showed that the expression of CD44V was significantly increased in 7 cases of CMM compared with 5 cases of CN (P<0.05). The positive rates of CD44V expression in 7 cases of CMM and 40 cases of UMM were 71.43% and 62.5%, respectively; The expression of CD44V in sclera-affected individuals was significantly higher than that in sclera-affected individuals (P=0.0105). The expression of CD44V in UMM was significantly negatively correlated with proliferation index (PI) and percentage of S-phase cells (SPF). P<0.01; P<0.05). The above indicates that the overexpression of CD44V is related to the occurrence of CMM and UMM, and related to the local infiltration of UMM.
