目的:本文研究了光动力学疗法对大鼠胶质瘤照射后单态氧(1O2)含量的影响。材料与方法∶取56只SD大鼠皮下注射C6胶质瘤细胞,待肿瘤直径达2cm,随机分成7组,每组8只,其中一组为对照组,于肿瘤内注射光敏剂血啉甲醚(HMME,10mg/kg),对照组未注射光敏剂.注射后给予光动力治疗,光动力学疗法以功率为150mW/cm2的He-Ne激光照射15min,将照射后大鼠分别于照射后即刻,15min,30min,60min,8h,24h取出肿瘤样本,液氮冷冻储存,真空冷冻抽干,研磨后用电子自旋磁共振机测量单态氧含量。结果∶在所选定的剂量下,照射后单态氧浓度逐渐增加,至60min时达到高峰,照射后8h以后单态氧含量显著减低。结论:光动力治疗作用对肿瘤的抑制途径之一是光动力治疗后单态氧的产生短时间内迅速升高,对肿瘤细胞起DNA光敏作用,作用于细胞膜,线粒体以及核酸,达到杀死肿瘤细胞目的。
OBJECTIVE: The effects of photodynamic therapy on the content of singlet oxygen (1O2) in rat glioma after irradiation were studied. MATERIALS AND METHODS: Sixty-six SD rats were injected subcutaneously with C6 glioma cells. When the tumor diameter reached 2 cm, they were randomly divided into 7 groups with 8 rats in each group. One group was control group. The photosensitizer, (HMME, 10 mg / kg), and no photosensitizer was injected into the control group.Photodynamic therapy was given after the injection, and He-Ne laser with a power of 150 mW / cm2 was applied for 15 minutes by photodynamic therapy. After irradiation, Tumor samples were removed immediately, 15 min, 30 min, 60 min, 8 h, 24 h later. The samples were frozen in liquid nitrogen and drained by vacuum freeze-drying. The content of singlet oxygen was measured by ESR after grinding. Results: At the selected dose, the concentration of singlet oxygen increased gradually after irradiation, reached the peak at 60 min and the content of singlet oxygen decreased significantly after 8 h. Conclusions: Photodynamic therapy is one of the ways to inhibit the tumor. After the photodynamic therapy, the production of singlet oxygen rapidly rises in a short period of time, plays a role of DNA photosensitizing effect on tumor cells, acting on the cell membrane, mitochondria and nucleic acid to kill the tumor The purpose of the cell.
