目的:研究纤体降脂Ⅰ号对大鼠去卵巢后肥胖的预防作用,探讨其预防肥胖的作用机制。方法:270 g左右的SD雌性大鼠70只,按体重随机选出假手术组10只,其余采用双侧去卵巢法,建立去卵巢大鼠模型。造模成功后,将大鼠随机分为模型对照组、纤体降脂Ⅰ号高剂量组(13.2 g.kg-1.d-1)、中剂量组(6.6 g.kg-1.d-1)、低剂量组(3.3 g.kg-1.d-1)、曲美组(8.9×10-4g.kg-1.d-1)、己烯雌酚组(2.2×10-5g.kg-1.d-1),开始灌胃给药干预,连续3周,实验结束后,从大鼠体重、腹脂、Lee’s指数、血脂等方面评价其药效;从血清雌二醇和瘦素含量以及下丘脑瘦素的表达,探讨其作用机制。结果:纤体降脂Ⅰ号能明显抑制模型鼠体重和食量的增长(P<0.05或P<0.01),降低腹脂含量和Lee’s指数(P<0.05或P<0.01),有效预防大鼠去卵巢后肥胖;显著降低大鼠血清甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)水平(P<0.05或P<0.01),有升高高密度脂蛋白胆固醇(HDL-C)的趋势;能明显提高大鼠血清雌二醇水平(P<0.05或P<0.01)。结论:纤体降脂Ⅰ号能够有效控制大鼠体重和食欲,预防大鼠去卵巢后肥胖的发生,降低血脂;提高血清雌激素含量,这可能是其预防大鼠去卵巢后肥胖的机制之一。
OBJECTIVE: To study the preventive effect of Soybean Preparation on the obesity after ovariectomy in ovariectomized rats, and to explore its mechanism of preventing obesity. Methods: Seventy-two female SD rats weighing 270 g were randomly divided into sham-operation group (n = 10) and ovariectomized rats (n = 10). After successful modeling, the rats were randomly divided into model control group, high-dose group (13.2 g.kg-1.d-1), medium dose group (6.6 g.kg- 1), low dose group (3.3 g.kg-1.d-1), trimetazidine group (8.9 × 10-4g.kg-1.d-1), diethylstilbestrol group (2.2 × 10-5g.kg-1 .d-1), began to intragastric administration intervention for 3 weeks, after the end of the experiment, the body weight, abdominal fat, Lee’s index, blood lipids and other aspects of evaluation of its efficacy; from the serum estradiol and leptin levels and The expression of leptin in thalamus and its mechanism of action. Results: Slimming lipid-lowering Ⅰ significantly inhibited body weight and food intake (P <0.05 or P <0.01), decreased abdominal fat content and Lee’s index (P <0.05 or P <0.01), and effectively prevented rats from going Ovarian obesity, significantly lower serum triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and total cholesterol (TC) levels in rats (P <0.05 or P <0.01), elevated high density lipoprotein Cholesterol (HDL-C) trend; can significantly increase serum estradiol levels (P <0.05 or P <0.01). Conclusion: Slimming lipid-lowering Ⅰ can effectively control body weight and appetite, prevent obesity after ovariectomy, lower blood lipids and increase serum estrogen levels, which may be the mechanism of preventing obesity after ovariectomy in rats one.
