利用正交试验确定了高压电脉冲电场和传统热水法提取海带多糖的方法的最佳工艺,并对2种工艺进行了比较,初步探讨高压脉冲的作用机理。结果表明,传统热水法提取海带多糖的最佳工艺为:海带粉与去离子水的料液比为1∶50,提取温度为60℃,提取时间为40 min。高压脉冲电场的最佳工艺为:海带粉与去离子水的料液比为1∶30,脉冲时间为20 min,脉冲电压为10 k V,脉冲频率为50 Hz。提取时间仅为传统热水法时间的1/2,海带多糖提取率比传统热水法提高45%。
The orthogonal experiment was used to determine the optimal process of high voltage pulse electric field extraction and traditional hot water extraction of kelp polysaccharide. The two processes were compared and the mechanism of high voltage pulse was discussed. The results showed that the optimum extraction process of kelp polysaccharides by traditional hot water method was as follows: the ratio of kelp powder to deionized water was 1:50, the extraction temperature was 60 ℃ and the extraction time was 40 min. The optimum process of high voltage pulsed electric field is as follows: the material-liquid ratio of kelp powder to deionized water is 1:30, the pulse time is 20 min, the pulse voltage is 10 kV and the pulse frequency is 50 Hz. The extraction time is only 1/2 of that of the traditional hot water method, and the extraction rate of kelp polysaccharide is 45% higher than that of the traditional hot water method.
