本文用3H-TdR(氚-胸腺嘧啶核苷)掺入的方法,检测了68例大肠癌、24例乳腺癌、20例卵巢癌肿瘤细胞的体外药物敏感性。结果显示,对大肠癌和乳腺癌以5-氟脲嘧啶(5-FU),丝裂霉素(MMC),环磷酰胺(CTX)的抑制率为高。且两药合用效果比单一药物效果好,三种以上药物联合使用可进一步提高疗效。卵巢癌的病例中,我们对每一组药物都作了亚甲蓝增敏试验,结果亚甲蓝增敏组的抑制率全部大于相应药物无亚甲蓝增敏组(P<0.01),提示亚甲蓝可以用作增敏剂提高化疗药的疗效。用本方法可筛选出对特定病人肿瘤灵敏的药物,具有简便、快速、有效、价廉的特点。
In this paper, the in vitro drug sensitivity of 68 cases of colorectal cancer, 24 cases of breast cancer, and 20 cases of ovarian cancer cells was detected by 3H-TdR (氚-thymidine) incorporation. The results showed that the inhibition rates of 5-fluorouracil (5-FU), mitomycin (MMC), and cyclophosphamide (CTX) were high in colorectal cancer and breast cancer. The effect of the combination of the two drugs is better than that of a single drug, and the combination of three or more drugs can further improve the curative effect. In the case of ovarian cancer, we made a methylene blue sensitization test for each group of drugs. The results showed that the inhibition rate of the methylene blue sensitization group was greater than that of the corresponding drug without methylene blue sensitization group (P<0.01). It suggests that methylene blue can be used as a sensitizer to improve the efficacy of chemotherapeutic agents. With this method, it is possible to screen out drugs that are sensitive to specific patients’ tumors, which are simple, rapid, effective, and inexpensive.
