目的研究2,3,7,8-四氯二苯并二噁英(2,3,7,8-tetrachlorodibenzo--pdioxin,TCDD)对大鼠血清中氧化应激指标的影响。方法清洁级Wistar雄性大鼠32只,体重(100±10)g,随机分成4组,连续经口染毒TCDD 90 d,剂量分别为2.5、25、250 ng/kg,对照组给予二甲基亚砜(DMSO),测定SOD、CuZn-SOD、Mn-SOD、GSH-Px、GST活性及MDA的含量。结果TCDD染毒90 d后,与对照组相比,各染毒组雄性大鼠血清中MDA含量明显增高(P<0.05);各染毒组总SOD、CuZn-SOD、MnSOD、GSH-Px活力均有所下降,其中除CuZn-SOD外,与对照组相比差异均有统计学意义(P<0.05)。同时各染毒组GST活力显著增加,且各组间差异有统计学意义(P<0.05)。结论亚慢性染毒引起MDA含量增加,大鼠血清中抗氧化指标出现异常改变,提示体内出现氧化与抗氧化系统失衡。
Objective To investigate the effect of 2,3,7,8-tetrachlorodibenzo-pdioxin (TCDD) on oxidative stress in serum of rats. Methods Thirty-two Wistar male rats were randomly divided into 4 groups (n = 10). The rats were continuously treated with oral TCDD for 90 days at doses of 2.5, 25 and 250 ng / kg, respectively. The control group was given dimethyl Sulfoxide (DMSO). The activities of SOD, CuZn-SOD, Mn-SOD, GSH-Px, GST and the contents of MDA were measured. Results Compared with the control group, the content of MDA in serum of male rats exposed to TCDD for 90 d was significantly higher than that of the control group (P <0.05). The activities of SOD, CuZn-SOD, MnSOD and GSH-Px (P <0.05), except CuZn-SOD, compared with the control group. At the same time, the activity of GST in each group increased significantly, and there was significant difference among the groups (P <0.05). Conclusion Sub-chronic exposure causes the increase of MDA content, and abnormal changes of antioxidant index in serum of rats, suggesting the imbalance of oxidation and anti-oxidation system in vivo.
