目的:探讨粒细胞集落刺激因子(G-CSF)动员正常供者外周血造血干细胞的效果、影响因素及不良反应。方法:对20例造血干细胞正常供者采用中位剂量G-CSF10.9μg/(kg·d)皮下注射3~5天后,使用COBESpectra血细胞分离机采集外周血干细胞。采用流式细胞仪检测采集物中CD34+细胞数。结果:第一次采集的单个核细胞(MNC,个)及CD34+细胞量(个)分别为(1.12~13.06,中位数3.10)×108/kg供者体重及(1.14~10.42,中位数3.8)×106/kg供者体重。MNC及CD34+细胞采集总量分别为(2.21~13.60,中位数5.80)×108/kg受者体重及(2.30~14.00,中位数6.40)×106/kg受者体重,均达到移植要求。男性CD34+细胞总量显著高于女性。不良反应较为轻微。供者年龄、体重、G-CSF剂量、采集前白细胞数等与采集所得MNC及CD34+细胞量无明显相关性。结论:G-CSF作为正常供者动员剂安全有效。
Objective: To investigate the effects, influencing factors and adverse reactions of granulocyte-colony stimulating factor (G-CSF) mobilizing peripheral blood hematopoietic stem cells from normal donors. Methods: 20 normal hematopoietic stem cell donors were injected subcutaneously with G-CSF (10.9 μg / (kg · d)) for 3 to 5 days. COBESpectra blood cells were used to collect peripheral blood stem cells. The number of CD34 + cells in the collected samples was detected by flow cytometry. Results: The numbers of mononuclear cells (MNCs) and CD34 + cells collected for the first time were (1.12 ~ 13.06, median 3.10) × 108 / kg and body weights of donors (1.14 ~ 10.42, 3.8) × 106 / kg donor body weight. The total body weight of MNC and CD34 + cells were (2.21 ~ 13.60, median 5.80) × 108 / kg body weight and body weight of recipients (2.30-14.00, median 6.40) × 106 / kg body weight, respectively. The total number of male CD34 + cells was significantly higher than that of women. Adverse reactions are mild. Donor age, body weight, G-CSF dose, number of white blood cells before collection and collection of MNC and CD34 + cells had no significant correlation. Conclusion: G-CSF is safe and effective as a normal donor.
