目的探讨牙本质基质蛋白-1(dentin matrix protein,DMP-1)对功能矫治前伸下颌过程中髁突骨软骨增殖的作用,为临床骨性Ⅱ类错矫形提供实验依据。方法选用4周龄健康雄性Sprague-Dawley(SD)大鼠70只,随机等量分为实验组和对照组,实验组大鼠全天佩戴下颌前伸斜面导板,对照组不做任何处理。于实验第1、3、7、14、21、28和35天每组分别处死大鼠5只,取双侧髁突,采用免疫组织化学方法及细胞图像分析系统检测DMP-1的表达变化。结果实验组大鼠髁突软骨中DMP-1表达于第3天开始增强,至第14天达到最高峰,强度-色调-饱和度(IHS)值(85.67±4.51)与对照组(10.67±1.53)相比差异有统计学意义(P<0.05)。结论 DMP-1参与大鼠下颌前伸过程髁突软骨适应性改建,促进髁突软骨增殖和软骨内成骨。
Objective To investigate the effect of dentin matrix protein (DMP-1) on the proliferation of condylar osteochondral cartilage during mandibular protraction and mandibular function, and to provide experimental evidence for the clinical skeletal Class Ⅱ malocclusion. Methods Seventy four-week-old healthy male Sprague-Dawley (SD) rats were randomly divided into experimental group and control group. Rats in experimental group were fitted with mandibular forward extending bevel guide all day without any treatment in control group. Five rats were killed in each group on the 1st, 3rd, 7th, 14th, 21st, 28th and 35th day respectively. The bilateral condyle was removed and the expression of DMP-1 was detected by immunohistochemistry and cell image analysis system. Results The expression of DMP-1 in the condylar cartilage increased from the third day to the peak on the 14th day. The intensity-intensity-saturation (IHS) value of the experimental group was 85.67 ± 4.51 and that of the control group was 10.67 ± 1.53 ) Compared with the difference was statistically significant (P <0.05). Conclusion DMP-1 is involved in condylar cartilage remodeling during mandibular protraction and promotes condylar cartilage proliferation and endochondral ossification.
