目的动态分析抗-HCV阳性献血者血浆miR-122的表达特点及与HCV感染诊断指标的关联。方法对133名抗-HCV阳性献血者、24名健康献血者及15名确证抗-HCV阳性追踪献血者血液标本抽提血浆miRNA,以实时荧光定量PCR法检测血浆miR-122,分析血浆miR-122相对表达量与HCV RNA、抗-HCV临界值指数(COI)等指标关联性。结果 ROC曲线分析:miR-122在抗-HCV+/HCV RNA+组和抗-HCV+/HCV RNA-组中诊断丙型肝炎的AUC(ROC曲线下面积)为0.830(95%CI:0.755-0.889),区分抗-HCV+/HCV RNA+组和抗-HCV+/HCV RNA-组的敏感性和特异性分别为74.7%和82.4%。追踪15例抗-HCV+/HCV RNA+献血者血浆miR-122表达明显降低(P<0.05)。结论血浆miR-122与HCV感染密切相关,确证HCV RNA感染献血者血浆miR-122随时间迁移表达降低。
Objective To analyze the expression of plasma miR-122 in anti-HCV positive blood donors and its correlation with the diagnostic index of HCV infection. Methods Plasma miRNAs were extracted from blood samples of 133 anti-HCV positive donors, 24 healthy donors and 15 confirmed anti-HCV positive blood donors. Real-time fluorescence quantitative PCR was used to detect plasma miR-122. Plasma miR- 122 relative expression level and HCV RNA, anti-HCV critical value index (COI) and other indicators. Results ROC curve analysis: The AUC (area under ROC curve) of miR-122 in diagnosing hepatitis C in anti-HCV + / HCV RNA + and anti-HCV + / HCV RNA- groups was 0.830 (95% CI: 0.755-0.889) The sensitivity and specificity for distinguishing anti-HCV + / HCV RNA + and anti-HCV + / HCV RNA- groups were 74.7% and 82.4%, respectively. The expression of miR-122 in plasma of 15 anti-HCV + / HCV RNA + blood donors was significantly lower (P <0.05). Conclusions Plasma miR-122 is closely related to HCV infection, confirming that the expression of plasma miR-122 in blood of HCV RNA-infected donors decreases with time.
