目的:研究维生素E琥珀酸酯(VES)诱导HER-2高表达乳腺癌细胞MDA-MB-453凋亡,以及Fas和TGF-β1表达的变化。方法:MTT法和Annexin V法分别用于检测不同浓度VES处理人乳腺癌MDA-MB-453细胞后生长情况和细胞凋亡情况,流式细胞术检测凋亡率;免疫组化比较凋亡前后Fas及TGF-β1表达变化。结果:不同浓度VES处理的细胞在不同时间均受到不同程度的抑制,随VES剂量的增加,抑制作用增强。其中20 mg/LVES处理组在24 h后受到明显抑制,第4天抑制率即接近100%;10 mg/L VES处理组7 d后细胞生长被抑制89.87%。经VES处理后,Fas及TGF-β1表达率差异有统计学意义,P<0.001。结论:VES可诱导HER-2高表达乳腺癌细胞MDA-MB-453凋亡,其机制可能与Fas和TGF-β1上调有关。
OBJECTIVE: To study the apoptosis of breast cancer cell line MDA-MB-453 induced by vitamin E succinate (VES) and the expression of Fas and TGF-β1 in HER-2-overexpressing breast cancer cells. Methods: MTT assay and Annexin V assay were used to detect the growth and apoptosis of human breast cancer MDA-MB-453 cells treated with different concentrations of VES, respectively. The apoptosis rate was detected by flow cytometry. Fas and TGF-β1 expression changes. Results: The cells treated with different concentrations of VES were inhibited to varying degrees at different times. With the increase of the dose of VES, the inhibition was enhanced. 20 mg / LVES treatment group was significantly inhibited after 24 h, the fourth day inhibition rate was close to 100%; 10 mg / L VES treatment group 7 d after inhibition of cell growth was 89.87%. After VES treatment, the expression of Fas and TGF-β1 showed significant difference (P <0.001). Conclusion: VES can induce the apoptosis of breast cancer cell line MDA-MB-453 with high expression of HER-2, which may be related to the up-regulation of Fas and TGF-β1.
