目的:研究蛇葡萄素钠(AMP-Na)协同卡铂对肺癌Lewis细胞增殖的抑制作用。方法:以6.25、12.5、25、50、100μg/ml AMP-Na+3.125、6.25、12.5、25、50μg/ml卡铂培养细胞4 h,测定细胞活力并计算抑制率与半数抑制浓度(IC50)。以12.5、25、50、100μg/ml AMP-Na+12.5μg/ml卡铂培养细胞12 h,流式细胞仪检测细胞半胱氨酸天冬氨酸蛋白水解酶3(Caspase-3)表达。结果:系列质量浓度AMP-Na与系列质量浓度卡铂联用后对细胞生长有明显抑制作用,随质量浓度的升高,卡铂对Lewis细胞的IC50逐渐降低;AMP-Na质量浓度范围在6.25~50μg/ml时,与3.125~25μg/ml卡铂联用后,对Lewis细胞增殖的协同抑制作用最强。AMP-Na与卡铂联合培养细胞12 h后,细胞Caspase-3表达明显增强。结论:AMP-Na与卡铂合用对Lewis细胞增殖具有协同抑制效应,其机制可能与AMP-Na激活细胞Caspase-3,从而诱导细胞凋亡有关。
Objective: To investigate the inhibitory effect of AMP-Na and carboplatin on the proliferation of Lewis lung cancer cells. Methods: The cells were cultured with 6.25,12.5,25,50,100μg / ml AMP-Na + 3.125,6.25,12.5,25,50μg / ml carboplatin for 4 hours, and the cell viability was measured and the inhibition rate and half-maximal inhibitory concentration (IC50) . The cells were cultured with 12.5, 25, 50, 100μg / ml AMP-Na + 12.5μg / ml carboplatin for 12 hours, and the expression of caspase-3 was detected by flow cytometry. Results: AMP-Na with series concentrations of carboplatin significantly inhibited the cell growth. With the increase of mass concentration, the IC50 of carboplatin on Lewis cells decreased gradually. The concentration of AMP-Na in the range of 6.25 ~ 50μg / ml, combined with 3.125 ~ 25μg / ml carboplatin, the synergistic inhibition of Lewis cell proliferation the strongest. After cultured with AMP-Na and carboplatin for 12 h, the expression of Caspase-3 was significantly increased. CONCLUSION: The combination of AMP-Na and carboplatin has a synergistic inhibitory effect on the proliferation of Lewis cells. The mechanism may be related to AMP-Na activating Caspase-3 and inducing cell apoptosis.
