视网膜移植研究的兴起为视网膜变性类疾病的治疗带来希望,由于供体缺乏、移植免疫等问题,视网膜色素上皮(RPE)细胞等的视网膜移植在临床上的应用发展受到阻碍。随着生物科学领域的发展,胚胎干细胞(ESCs)的特征鉴定及体外培养等技术的不断提高,ESCs可在体外诱导分化成RPE细胞,成为视网膜移植一大供体来源。本文就ESCs的体外分离培养、鉴定、诱导分化的研究及其在RPE移植中的应用研究作一综述。
The rise of retinal transplantation research brings hope for the treatment of retinal degenerative diseases. The development of retinal transplantation of retinal pigment epithelium (RPE) cells and the like has been hindered in clinical application because of lack of donor and transplantation immunity. With the development of biological sciences, the characteristics of embryonic stem cells (ESCs) and the continuous improvement of in vitro culture, ESCs can differentiate into RPE cells in vitro and become a source of retinal transplantation. In this paper, in vitro isolation and culture of ESCs, identification, induction of differentiation and its application in RPE transplantation are reviewed.
