目的:采用高效液相色谱法(HPLC)测定鸡树条荚蒾生长期不同部位绿原酸含量,为其资源合理利用提供科学依据。方法:采用VP-ODS C18(4.6mm×150mm,5μm)色谱柱,以乙腈∶0.4%磷酸(17∶83)为流动相,流速为1.0mL·min-1,检测波长为327nm。果实和叶片的绿原酸提取液经0.22μm滤膜过滤后直接进样,进样量为20μL。结果:绿原酸浓度在0.01~54.0μg/mL范围内与峰面积呈良好的线性关系(r=0.999 7)。8月果实中绿原酸含量最高,为49.35μg/mL。结论:鸡树条荚蒾生长期果实中绿原酸含量变化显著,且8月份积累最多。
OBJECTIVE: To determine the content of chlorogenic acid in different parts of the growth stage of Viburnum chinense by HPLC with high performance liquid chromatography (HPLC), and provide a scientific basis for its rational utilization of resources. Methods: The mobile phase consisted of VP-ODS C18 (4.6 mm × 150 mm, 5 μm) with acetonitrile: 0.4% phosphoric acid (17:83) at a flow rate of 1.0 mL · min-1 and a detection wavelength of 327 nm. The chlorogenic acid extract of fruits and leaves were directly injected through a 0.22 μm membrane filter, and the injection volume was 20 μL. Results: The concentration of chlorogenic acid in the range of 0.01 ~ 54.0μg / mL showed a good linear relationship with the peak area (r = 0.999 7). The highest content of chlorogenic acid in fruits in August was 49.35μg / mL. Conclusion: The content of chlorogenic acid in the fruit of Viburnum chingii var. Chinensis was significantly changed, and accumulated most in August.
