目的探讨低分子肝素钙(LMWH)对急性下肢深静脉血栓(DVT)大鼠血清白介素6(IL-6)和肿瘤坏死因子α(TNF-α)水平的影响。方法将57只SD大鼠随机分为空白对照组20只和DVT组37只,DVT组予以结扎近心端股静脉,空白对照组大鼠不予结扎。再将DVT组大鼠随机分为实验对照组18只和LMWH组19只。空白对照组和实验对照组大鼠均给予2 m L生理盐水皮下注射,LMWH组大鼠给予2 m L LMWH皮下注射,每12小时注射1次,60 h后停止注射。分别于术前、术后36 h和术后72 h测量3组大鼠左下肢膝关节上6 mm处的左下肢周径及血清IL-6和TNF-α水平,并进行组间比较。结果术前空白对照组、实验对照组及LMWH组的左下肢周径、血清IL-6水平和血清TNF-α水平比较差异均无统计学意义(P>0.050),但术后36 h和术后72 h 3组的左下肢周径、血清IL-6水平和血清TNF-α水平比较差异均有统计学意义(P<0.050),均是实验对照组>LMWH组>空白对照组。结论急性下肢DVT SD大鼠行LMWH治疗后其血清IL-6和TNF-α水平均明显降低,提示LMWH对急性下肢DVT具有抗炎作用。
Objective To investigate the effects of low molecular weight heparin (LMWH) on serum levels of interleukin 6 (IL-6) and tumor necrosis factor-α (TNF-α) in acute lower extremity deep vein thrombosis (DVT) Methods Totally 57 SD rats were randomly divided into blank control group (n = 20) and DVT group (n = 37). The DVT group was ligated with the proximal femoral vein and the blank control group was not ligated. Then the rats in DVT group were randomly divided into experimental control group (n = 18) and LMWH group (n = 19). The rats in the blank control group and the experimental control group were injected subcutaneously with 2 ml normal saline. Rats in the LMWH group were injected subcutaneously with 2 ml LMWH once every 12 hours and stopped 60 hours later. The left lower extremity circumference, serum IL-6 and TNF-α levels at 6 mm in the left lower extremity knee joint of the three groups were measured before operation, 36 h after operation and 72 h after operation respectively. Results There were no significant differences in the left lower extremity circumference, serum IL-6 level and serum TNF-α level between preoperative blank control group, experimental control group and LMWH group (P> 0.050) The left lower limb circumference, serum IL-6 level and serum TNF-α level in the three groups at 72 h after operation were significantly different (P <0.050), both of which were experimental control group> LMWH group> blank control group. Conclusion The serum levels of IL-6 and TNF-α in the lower extremity DVT SD rats after LMWH treatment are significantly lower, suggesting that LMWH has an anti-inflammatory effect on acute extremity DVT.
