目的 研究血小板反应蛋白 1(TSP 1)和血管内皮生长因子(VEGF)的平衡在肝细胞 癌(HCC)新生血管生成中的作用。方法 采用免疫组织化学SABC法检测37例HCC及其癌旁肝 组织和6例正常肝组织中TSP 1、VEGF和CD34的表达情况,研究TSP 1和VEGF的表达与HCC 的临床病理资料及新生血管生成的关系。结果 37例HCC中TSP 1阳性率为62.2%(23/37),低 于相应的癌旁肝组织和正常肝组织(P<0.05)。HCC中VEGF的阳性率为97.3%(36/37),高于相 应的癌旁肝组织和正常肝组织(P<0.05)。有镜下静脉浸润的HCC组织中TSP 1的表达低于无镜 下静脉浸润者,而VEGF的表达则高于无镜下静脉浸润者(P<0.05)。等级相关分析显示,HCC中 VEGF的表达与CD34的表达呈正相关关系,(rs=0.333,P<0.05),而TSP 1的表达与CD34的表达 则呈负相关关系,相关系数(rs=-0.393,P<0.05)。结论 TSP 1低表达与VEGF过表达之间的 不平衡性是决定HCC组织中新生血管生成的关键因素之一。
Objective To investigate the role of balance of thrombospondin 1 (TSP 1) and vascular endothelial growth factor (VEGF) in angiogenesis of hepatocellular carcinoma (HCC). Methods Immunohistochemical SABC method was used to detect the expression of TSP 1, VEGF and CD34 in 37 cases of HCC, adjacent normal liver tissue and 6 cases of normal liver tissue. The expressions of TSP 1 and VEGF and the clinicopathological data of HCC and neovascularization Generated relationship. Results The positive rate of TSP 1 in 37 cases of HCC was 62.2% (23/37), which was lower than that of the corresponding paracancerous liver tissue and normal liver tissue (P <0.05). The positive rate of VEGF in HCC was 97.3% (36/37), which was higher than that in the corresponding paracancer liver tissue and normal liver tissue (P <0.05). The expression of TSP 1 in HCC tissues with microscopic vein infiltration was lower than that in those without infiltration, while the VEGF expression was higher than those without infiltration of the vein (P <0.05). Hierarchical correlation analysis showed that there was a positive correlation between the expression of VEGF and the expression of CD34 in HCC (rs = 0.333, P <0.05), while the expression of TSP 1 was negatively correlated with the expression of CD34 (rs = -0.393 , P <0.05). Conclusion The imbalance between low expression of TSP 1 and overexpression of VEGF is one of the key factors that determine neovascularization in HCC tissues.
