用NADPH-黄递酶(NADPH-d)组织化学结合免疫细胞化学双标记方法,对大鼠部分脑区内一氧化氮合酶(NOS)分别与5-羟色胺(5-HT)、酪氨酸羟化酶(TH)和乙酰胆碱(ACh)等共存神经元进行了观察。结果表明在中缝背核(DR)背外侧区,绝大多数NOS神经元与5-HT共存,但5-HT神经元仍有一半以上未表现出NADPH-d阳性。在脑桥蓝斑(LC)腹内侧部存在大量NOS神经元,其中一小部分与去甲肾上腺素(NA)能神经元(以TH为标记)共存。在中脑黑质(SN)区观察到少量NOS神经元散在地分布干大量的多巴胺(DA)能神经元(以TH为标记)之中,此区还存在大量NADPH-d阳性纤维,但没有观察到NOS与DA共存神经元。在前脑中央隔区(MS)、Broca斜带核水平支(hDB)和垂直支(vDB),92%以上的NOS神经元与ACh共存,这些共存神经元约占所有胆碱能神经元的42%。纹状体内也有不少NOS神经元,但它们不与ACh共存。上述NOS分别与5-HT、NA和ACh等经典神经递质共存的结果,为NO参与多种递质功能的调节提供了形态学基础。
NADPH-d histochemical and immunocytochemical double labeling method was used to detect the expression of nitric oxide synthase (NOS) in some brain regions of rats with 5-hydroxytryptamine (5-HT), tyrosine Coexist neurons such as hydroxylase (TH) and acetylcholine (ACh) were observed. The results showed that most of the NOS neurons co-existed with 5-HT in the dorsolateral region of the dorsal raphe nucleus (DR), but more than half of 5-HT neurons did not show positive NADPH-d. A large number of NOS neurons exist in the ventral part of the locus locus coeruleus (LC), and a small part of them coexist with noradrenaline (NA) - capable neurons (labeled with TH). A small number of NOS neurons scattered in the midbrain substantia nigra (SN) area were scattered among a large number of dopaminergic neurons (labeled with TH), which also contained a large number of NADPH-d-positive fibers but no Neurons coexisting with NOS and DA were observed. More than 92% of NOS neurons coexist with ACh in the basal forebrain (MS), BrDB (hDB) and vertical branch (vDB), and these coexisting neurons account for about 80% of all cholinergic neurons 42%. There are many NOS neurons in the striatum, but they do not coexist with ACh. The coexistence of these NOS and classical neurotransmitters such as 5-HT, NA and ACh, respectively, provided the morphological basis for NO’s involvement in the regulation of various neurotransmitter functions.
