[目的]菌株NJ13是1株对人参常见病原菌有较强拮抗作用的内生细菌,为了有效防治人参病害,进行了菌种鉴定和发酵培养工艺的试验。[方法]通过培养特性、形态特征、生理生化特性和16S rDNA同源性序列分析,对菌株NJ13进行鉴定;采用牛津杯法测定抗菌谱;同时通过发酵单因素实验和各因素正交试验确定菌株的发酵工艺。[结果]确定菌株NJ13为甲基营养型芽孢杆菌(Bacillus methylotrophicus),该菌对多种植物病原菌有拮抗作用,适宜的发酵培养基配方:葡萄糖3%,淀粉1.5%,酵母浸粉1.5%,K2HPO40.1%,NaCl 0.5%;最适培养条件:接种量4%,装液量75 mL/250mL,起始pH值8.0,28℃、180 r/min摇瓶培养96 h。[结论]为人参病害的防治提供新的生物防治资源。
[Objective] The strain NJ13 was an endophytic bacterium which had a strong antagonism against the common pathogen of ginseng. In order to effectively prevent and cure the disease of ginseng, the strain NJ13 was tested for its strain identification and fermentation culture process. [Method] The strain NJ13 was identified through the culture characteristics, morphological characteristics, physiological and biochemical characteristics and 16S rDNA sequence homology analysis. The antibacterial spectrum of strain NJ13 was determined by the Oxford cup method. The single strain factor and the orthogonal test were used to determine the strain The fermentation process. [Result] The strain NJ13 was confirmed to be Bacillus methylotrophicus, which antagonized many plant pathogenic bacteria. The suitable fermentation medium formula was glucose 3%, starch 1.5%, yeast extract 1.5% K2HPO40.1% and NaCl 0.5%. The optimal culture conditions were inoculation amount of 4%, liquid volume of 75 mL / 250 mL, initial pH of 8.0, 28 ℃ and shaking culture at 180 r / min for 96 h. [Conclusion] This research provided new biological control resources for the prevention and treatment of ginseng diseases.
