目的:建立复方金银花颗粒高效液相特征图谱的测定方法,并对其中绿原酸、黄芩苷和连翘苷3种指标性成分进行含量测定。方法:采用Phenomenex C18色谱柱(4.6 mm×250 mm,4μm),流动相乙腈-0.4%磷酸溶液梯度洗脱,流速1 mL.min-1,柱温35℃,检测波长228 nm,记录时间80 min,对5批同一厂家复方金银花颗粒的特征图谱进行研究及含量测定。结果:初步建立了复方金银花颗粒的高效液相特征图谱,标定了13个共有特征峰。各共有峰相对保留时间的RSD<4%,相对峰面积的RSD<4%。待测的3种成分均达到基线分离,绿原酸、黄芩苷和连翘苷的线性范围分别为0.11~5.57μg(r=0.999 9),0.05~2.66μg(r=1),0.01~0.52μg(r=1),回收率分别为98.9%,100.3%,99.2%。结论:方法稳定、准确、有效,可以全面的反映复方金银花颗粒的质量,可作为控制复方金银花颗粒质量的依据。
OBJECTIVE: To establish a method for the determination of the HPLC fingerprint of compound honeysuckle, and to determine the content of three index components of chlorogenic acid, baicalin and forsythin. METHODS: The mobile phase was eluted with a mobile phase of acetonitrile-0.4% phosphoric acid with a gradient of 1 mL.min-1 on a Phenomenex C18 column (4.6 mm × 250 mm, 4 μm) at a detection temperature of 228 nm and a recording time of 80 nm min, the five batches of the same manufacturer compound honeysuckle particles map of the characteristics and determination. Results: The high performance liquid chromatogram of compound honeysuckle particles was initially established and 13 common characteristic peaks were calibrated. The relative retention time of each common peak RSD <4%, relative peak area RSD <4%. The linear range of chlorogenic acid, baicalin and forsythin were 0.11-5.57μg (r = 0.999 9), 0.05-2.66μg (r = 1), 0.01-0.52 μg (r = 1), the recoveries were 98.9%, 100.3% and 99.2%, respectively. Conclusion: The method is stable, accurate and effective, which can fully reflect the quality of compound honeysuckle particles and can be used as the basis for controlling the quality of compound honeysuckle particles.
