目的观察葡萄柚漱口液对不同阶段变异链球菌生物膜的作用效果,并与西吡氯铵漱口液相比较。方法体外形成3、24、48 h变异链球菌生物膜模型,各组漱口液作用1 min后,(葡萄柚漱口液为实验组,设PBS液和西吡氯铵漱口液作为空白和阳性对照组),荧光染料染色生物膜中的死菌和活菌,激光共聚焦扫描显微镜观察并比较漱口液处理前后活菌百分比的变化。结果葡萄柚漱口液与空白组相较,各时段生物膜经处理后,3 h变异链球菌生物膜外层和中间层的活菌百分比,以及24、48 h生物膜外层的活菌百分比均有明显降低,两者间均有显著性差异(P<0.05);与阳性组相比较,二者间各层活菌百分比无显著性差异(P>0.05)。结论葡萄柚漱口液对各时段生物膜的外层细菌有杀伤作用,但仅能影响早期(3 h)变异链球菌生物膜的深层细菌生存,与西吡氯铵漱口液对变异链球菌生物膜的作用效果相似。
Objective To observe the effect of grapefruit gargle on the biofilm of Streptococcus mutans at different stages and compare it with cetylpyridinium chloride mouthwash. Methods Streptococcus mutans biofilm models were established at 3, 24 and 48 h in vitro. After the mouthwash was applied for 1 min in each group (grapefruit mouthwash was used as experimental group, PBS and cetylpyridinium chloride as blank and Positive control group). The dead bacteria and viable cells in the biofilm were stained with fluorescent dye. The change of the percentage of viable cells before and after treatment with gargle was observed by laser confocal scanning microscope. Results Compared with the blank group, the percentage of viable cells in the outer layer and the middle layer of Streptococcus mutans biofilm at 3 h and the percentage of viable cells in the outer layer of biofilm at 24 and 48 h were (P <0.05). Compared with the positive group, the percentage of viable cells in each layer had no significant difference (P> 0.05). Conclusions Grapefruit mouthwash has a killing effect on the outer layer of biofilm in each period, but it can only affect the deep-seated bacterial survival of the Streptococcus mutans biofilm in early stage (3 h) Biofilm effect similar.
