研究了12例健康人和41例甲亢患者血清中TS-H和球蛋白的相互作用。 方法:用Chopra等法制备血清。用双缩脲反应测定样品的蛋白质浓度。用三种方法试验球蛋白制品的甲状腺刺激免疫球蛋白(TSI)活性:(1)按Orgiazzi等法测定人甲状腺腺苷酸环化酶的刺激作用;(2)按Onaya等法测定胶质微滴的形成;(3)按Smith等法用人甲状腺细胞膜进行放射受体分析。使腺苷酸环化酶活性和胶质微滴形成增加40%以上的,或将甲状腺细胞膜上的TSH取代30%以上的样品作为TSI阳性。无这些种活性的健康人的样品作为TSI阴性。
The interaction between TS-H and globulin in serum of 12 healthy subjects and 41 hyperthyroid patients was studied. Methods: Chopra and other methods to prepare serum. The biuret reaction was used to determine the protein concentration of the sample. Thyroid stimulating immunoglobulin (TSI) activity of globulin preparations was tested in three ways: (1) stimulation of human thyroid adenylate cyclase by Orgiazzi et al., And (2) glial microanalysis as determined by Onaya et al. Drop formation; (3) by Smith and other methods using thyroid cell membrane radioreceptor analysis. Increases adenylyl cyclase activity and glial droplets formation by more than 40%, or TSH substitution in thyroid cell membranes by more than 30% of samples as TSI positive. Samples without these active, healthy individuals were negative for TSI.
