目的探讨瑞香素对CIA大鼠脾T淋巴细胞杀瘤效应的影响。方法建立胶原诱导关节炎(CIA)大鼠模型。大鼠随机分为健康对照组、CIA模型对照组、瑞香素治疗组、阳性药物对照组,连续治疗28天后处死大鼠,分离脾细胞。MTT法测定Con A诱导大鼠脾T淋巴细胞增殖率。MTT法检测大鼠脾T淋巴细胞对瘤细胞(肝癌细胞株SMMC-7721)的杀伤率。结果 1瑞香素治疗组脾T淋巴细胞增殖率为(161.11±14.05)%,与CIA模型对照组(55.61±10.88)%、阳性药物对照组(13.00±12.14)%、健康对照组(101.35±16.79)%比均明显增高(P<0.05)。2瑞香素治疗组大鼠脾T淋巴细胞对瘤细胞杀伤率为(23.50±11.08)%,与CIA模型对照组(15.89±8.77)%、阳性药物对照组(7.74±8.84)%比均明显增高(P<0.05),接近健康对照组(24.35±13.45)%。结论瑞香素能明显提高CIA大鼠脾T淋巴细胞增殖率和脾T淋巴细胞对瘤细胞杀伤率,有效促进CIA大鼠免疫功能。
Objective To investigate the effect of daphnetin on the cytotoxicity of splenic T lymphocytes in CIA rats. Methods Collagen-induced arthritis (CIA) rat model was established. The rats were randomly divided into healthy control group, CIA model control group, daphnetin treatment group and positive drug control group. After continuous treatment for 28 days, rats were killed and splenocytes were isolated. ConA induced proliferation rate of spleen T lymphocytes in rats by MTT method. The killing rate of rat splenic T lymphocytes to tumor cells (hepatocellular carcinoma cell line SMMC-7721) was determined by MTT assay. Results The proliferation rate of splenic T lymphocytes in Daphnetin-treated group was (161.11 ± 14.05)%, which was significantly higher than that in CIA model control group (55.61 ± 10.88)%, positive drug control group (13.00 ± 12.14)% and healthy control group (101.35 ± 16.79 )% Were significantly higher (P <0.05). The killing rate of splenic T lymphocytes to the tumor cells in the daphnetin-treated rats was (23.50 ± 11.08)%, which was significantly higher than that in the CIA model control group (15.89 ± 8.77)% and the positive drug control group (7.74 ± 8.84)% (P <0.05), close to the healthy control group (24.35 ± 13.45)%. Conclusion Daphnetin can significantly increase the proliferation rate of spleen T lymphocytes and the killing rate of splenic T lymphocytes to tumor cells in CIA rats, and effectively promote the immune function of CIA rats.
