目的探讨大豆甙元对子宫内膜癌细胞增殖的影响及其作用机制,为临床应用植物雌激素提供理论依据。方法体外培养子宫内膜癌Ishikawa细胞,采用MTT法检测不同浓度大豆甙元对子宫内膜癌细胞增殖的影响,同时采用RT-PCR及Western Blot方法检测细胞中雌激素受体相关受体α(ERRα)mRNA及蛋白水平的变化。结果大豆甙元对Ishikawa细胞体外增殖有明显的抑制作用,并随药物浓度的增加及作用时间的延长其抑制作用逐渐增加,呈剂量及时间依赖性。低浓度(<40 mmol/L)大豆甙元对细胞中ERRαmRNA及蛋白水平无明显影响,而高浓度(≥40 mmol/L)大豆甙元可上调细胞中ERRα的表达。结论大豆甙元可抑制子宫内膜癌细胞的增殖,这种抑制作用可能通过调控细胞中ERRα的表达实现。
Objective To investigate the effect of daidzein on the proliferation of endometrial carcinoma cells and its mechanism of action, and provide a theoretical basis for the clinical application of phytoestrogens. Methods Ishikawa cells were cultured in vitro. MTT assay was used to detect the effect of daidzein on the proliferation of endometrial carcinoma cells. RT-PCR and Western Blot were used to detect the expression of estrogen receptor-related receptor alpha ( ERRα) mRNA and protein levels. Results Daidzein significantly inhibited the proliferation of Ishikawa cells in vitro. With the increase of drug concentration and prolongation of action time, daidzein increased its inhibitory effect in a dose and time dependent manner. Low concentration (<40 mmol / L) daidzein had no significant effect on ERRαmRNA and protein levels, while high concentration (≥40 mmol / L) daidzein increased the expression of ERRα in cells. Conclusion Daidzein inhibits the proliferation of endometrial carcinoma cells. This inhibitory effect may be through the regulation of ERRα expression in cells.
