目的:探讨离子霉素对Jurkat T细胞增殖的抑制作用。方法:以Jurkat T细胞为模型,与5种细胞增殖抑制剂比较,观察离子霉素对Jurkat T细胞集落形成的影响,分析不同浓度离子霉素对Jurkat T细胞增殖的影响;分析离子霉素对JurkatT细胞周期的影响;分析离子霉素对Jurkat T细胞凋亡的作用。本实验结果用统计软件包SPSS10.0进行处理,数据以均数±标准差(x±s)表示,多组间比较采用One-Way ANOVA,两组间比较用非配对Student’st检验。结果:随着离子霉素浓度从0.1mg/L逐渐增至4.0mg/L,JurkatT细胞的增殖逐渐减弱,以4.0mg/L离子霉素的抑制作用最为明显,呈剂量依赖关系,且该浓度下,离子霉素对Jurkat T细胞增殖的抑制作用较PD98059、AG490、GF109203X和Genistein明显。离子霉素可使Jurkat T细胞周期停滞于G0/G1期,阻止其进入S期并可促进JurkatT细胞的凋亡。结论:离子霉素对Jurkat T细胞增殖有明显的抑制作用,可使Jurkat T细胞周期停滞于G0/G1期,阻止其进入S期,且对Jurkat T细胞凋亡有显著促进作用。
Objective: To investigate the inhibitory effect of ionomycin on the proliferation of Jurkat T cells. METHODS: Jurkat T cells were used as a model to compare the effects of ionomycin on Jurkat T cell colony formation and the effects of different concentrations of ionomycin on the proliferation of Jurkat T cells. The effects of ionomycin on JurkatT cell cycle; analysis of ionomycin on Jurkat T cell apoptosis. The experimental results using statistical software package SPSS10.0 for processing, the data as mean ± standard deviation (x ± s) that said one-way ANOVA comparison between groups, unpaired Student’s test between the two groups. Results: As the concentration of ionomycin gradually increased from 0.1mg / L to 4.0mg / L, the proliferation of Jurkat T cells gradually weakened, and the inhibitory effect of ionomycin at the concentration of 4.0mg / L was most obvious in a dose-dependent manner. , The inhibitory effect of ionomycin on Jurkat T cell proliferation was more obvious than that of PD98059, AG490, GF109203X and Genistein. Ionomycin can arrest the Jurkat T cell cycle in the G0 / G1 phase, prevent it from entering the S phase and promote the apoptosis of Jurkat T cells. CONCLUSION: Ionomycin can significantly inhibit the proliferation of Jurkat T cells, arresting the cycle of Jurkat T cells in G0 / G1 phase, preventing them from entering S phase and promoting the apoptosis of Jurkat T cells.
