目的:筛选并克隆人肝细胞cDNA文库中与乙型肝炎病毒(HBV)前-前-S蛋白相互作用蛋白的基因,探讨前-前- S在HBV致病中的作用. 方法:构建前-前-S的酵母细胞表达载体,采用酵母双杂交系统筛选人肝细胞cDNA文库,利用核苷酸数据库及生物信息学技术,对于筛选结果进行分析. 结果:获得了54个与前-前-S蛋白特异性结合的阳性克隆,其中包括30种已知蛋白基因和8个未知功能基因. 结论:克隆出与乙型肝炎病毒前-前-S蛋白结合的肝细胞蛋白基因,为进一步研究前-前-S在HBV致病中的作用奠定了基础.
Objective: To screen and clone the gene of hepatitis B virus (HBV) pre-S protein interacting with human hepatocyte cDNA library and to explore the role of pre-pre-S in the pathogenesis of HBV.Methods: Pre-S yeast cell expression vector, the yeast two-hybrid system screening of human hepatocyte cDNA library, the use of nucleotide database and bioinformatics analysis of the screening results.Results: 54 were obtained with pre-pre-S Including 30 known and 8 unknown functional genes.Conclusion: Hepatocyte protein gene which binds to hepatitis B virus pre-pre-S protein is cloned, Pre-S in the pathogenesis of HBV laid the foundation.
