目的 :探讨健脾益气方 (JPYQR)抑制胃粘膜损伤的作用机理。方法 :采用束缚水浸法造成大鼠急性胃粘膜损伤 ,测定溃疡指数 ,检测血浆及胃组织中热休克蛋白 (heatshockprotein ,HSP70 )的变化 ,并对胃组织细胞HSPs进行免疫组织化学研究。结果 :用中药JPYQR和西药丽珠得乐预处理的大鼠 ,溃疡指数小于模型组 (P <0 0 0 1 ) ,而JPYQR组又小于丽珠得乐组 (P <0 0 1 ) ;预处理两组血浆及胃组织中HSP70 水平相反明显高于模型组 (P <0 0 5 ) ,而JPYQR组血浆中HSP70 水平又显著高于丽珠得乐组 (P <0 0 5 ) ,二者胃组织中的HSP70 水平虽有差异 ,但无统计学意义 (P >0 0 5 ) ;免疫组织化学研究提示腺体表达大量HSP70 和HSP60 ,按模型组、丽珠得乐组、JPYQR组依次增强。结论 :中药健脾益气方能诱导大鼠提高HSPs的表达 ,防止胃粘膜损伤 ,HSPs可能介导胃粘膜防御。
Objective: To explore the mechanism of inhibiting gastric mucosal damage by JPYQR. METHODS: Acute gastric mucosal injury was induced by irreversible water immersion in rats. Ulcer index was measured. Changes of heat shock protein (HSP70) in plasma and stomach tissue were detected. Immunohistochemical study of HSPs in gastric tissue was performed. RESULTS: Rats pretreated with traditional Chinese medicine JPYQR and western medicine Livzondele had lower ulcer index than the model group (P <0 01), while JPYQR group was smaller than Livzondele group (P <0 01); The HSP70 levels in the plasma and stomach tissues of the two groups were significantly higher than those in the model group (P < 0.05), while the plasma HSP70 levels in the JPYQR group were significantly higher than those in the Livzondele group (P <0 05). Although the level of HSP70 in gastric tissue was different, it was not statistically significant (P > 0.05). Immunohistochemical studies revealed that the gland expresses a large amount of HSP70 and HSP60, followed by the enhancement of the model group, the Livzon Dele group, and the JPYQR group. . Conclusion: The Chinese medicine Jianpiyiqi can induce the expression of HSPs in rats and prevent gastric mucosal injury. HSPs may mediate gastric mucosal defense.
