目的:初步探讨胃泌素是否通过STAT3信号通路调控大肠癌细胞的增殖,旨在明确胃泌素调控大肠癌细胞增殖的分子机制。方法:体外培养的SW480细胞同步化疗后加入各实验组药物培养72h,流式细胞仪检测各组药物作用下细胞增殖指数的变化,RT-PCR方法检测各实验组细胞内STAT3mRNA表达水平的变化。结果:5肽胃泌素组能通过上调STAT3mRNA的表达而促进SW480的增殖,流式细胞术结果显示,胃泌素组细胞增殖指数为(37.54±5.17)%较对照组(27.72±5.00)%明显上升,P<0.05;在mRNA水平,胃泌素组SW480细胞STAT3mRNA基因相对表达为(0.68±0.087)也显著高于对照组(0.44±0.047),P<0.05。结论:STAT3可能是胃泌素促进大肠癌增殖的一个下游靶点,胃泌素可能通过STAT3信号通路调控大肠癌的增殖。
OBJECTIVE: To investigate whether gastrin regulates the proliferation of colorectal cancer cells through STAT3 signaling pathway and aims to clarify the molecular mechanism of gastrin in regulating the proliferation of colorectal cancer cells. Methods: SW480 cells cultured in vitro were added into the experimental groups after 72 hours. The proliferation of SW480 cells was detected by flow cytometry. The expression of STAT3 mRNA in each experimental group was detected by RT-PCR. Results: Gastrin group could promote the proliferation of SW480 by upregulating the expression of STAT3 mRNA. Flow cytometry showed that the proliferation index of gastrin group was (37.54 ± 5.17)% higher than that of control group (27.72 ± 5.00)% (P <0.05). At the mRNA level, STAT3 mRNA expression in SW480 cells in gastrin group was significantly higher than that in control group (0.68 ± 0.087, P <0.05). Conclusion: STAT3 may be a downstream target of gastrin to promote the proliferation of colorectal cancer. Gastrin may regulate the proliferation of colorectal cancer through STAT3 signaling pathway.
