取经福氏痢疾杆菌免疫后的Wistar大鼠,老龄鼠(20个月)10只,壮龄鼠(8个月)11只,雌雄兼用。通过腹膜腔注入致病量福氏痢疾杆菌F1b,24h后,处死取材;常规脱水,低度石蜡包埋、切片;ABC法染色,DAB显色。光镜下观察下丘脑弓状核促肾上腺皮质激素样免疫反应(ACTH-ir)和β-内啡肽样免疫反应(β-END-ir)阳性细胞的形态、分布和计数。图像分析仪测其灰度值。结果:光镜下计数下丘脑弓状核ACTH-ir和β-END-ir阳性细胞,老龄鼠明显低于壮龄鼠。多数鼠第三脑室室周层出现许多β-END-ir强阳性细胞。再感染老龄鼠弓状核ACTH-ir阳性细胞灰度值显著高于再感染壮龄鼠;其β-END-ir阳性细胞灰度值显著低于再感染壮龄鼠。提示:再感染老龄鼠虽经人工主动免疫,当受细菌攻击时,有呈高ACTH低β-END趋势;或许与其免疫功能衰退密切相关。
Wistar rats immunized with Shigella flexneri were enrolled. Ten old mice (20 months) and 11 young rats (8 months old) were used in both sexes. Pathogenic amount of Shigella flexneri was inoculated through the peritoneal cavity F1b, 24h later, sacrificed; routine dehydration, low paraffin embedding, sectioning; ABC staining, DAB color. The morphology, distribution and count of ACTH-ir and β-END-ir positive cells in the arcuate nucleus of the hypothalamus were observed under light microscope. Image analyzer to measure its gray value. Results: ACTH-ir and β-END-ir positive cells in the arcuate nucleus of the hypothalamus were counted under light microscope. The aged mice were significantly lower than the young mice. Most rats in the third ventricle peritoneal many β-END-ir strong positive cells. The gray value of ACTH-ir positive cells in arcuate nucleus of re-infected aged rats was significantly higher than that of re-infected young rats; the gray value of β-END-ir positive cells was significantly lower than that of re-infected young rats. Tip: Re-infected old mice, though artificially self-immunized, have a tendency of high ACTH low β-END when challenged by bacteria; and may be closely related to their decline in immune function.
