用电子显微镜直接观察大豆花叶病叶片压出液的稀释液图片,看出这一病毒是600—700×13—15mμ的线杆状粒子。根据稀释度估计病毒粒子在病叶组织中的浓度很低。这种方法在试验工作中用于直接诊断这一病害是有可能的。移去叶片的叶绿素,水解核酸后用光谱分析法测定大豆花叶病毒样品的光谱吸收曲线。健叶与病叶之间在吸收曲线的峯形上无特殊区别,在核酸含量的差别上也无明显的规律性。
Direct observation with electron microscopy of soybean mosaic leaf fluid exudate picture shows that the virus is 600-700 × 13-15mμ rod-shaped particles. The dilution of virus particles in diseased tissue is estimated to be low based on dilution. This method is possible for direct diagnosis of this disease in experimental work. The leaf chlorophyll was removed and the nucleic acid was hydrolyzed to determine the spectral absorption curve of soybean mosaic virus by spectroscopic analysis. There is no special difference in the peak shape of absorption curve between healthy leaves and diseased leaves, and there is no obvious regularity in the difference of nucleic acid content.
