目的:探讨理气消瘿片对实验性大鼠甲状腺肿的作用机制。方法:白色封闭群大鼠(SD),随机分为6组:正常对照组,模型组,理气消瘿片低、中、高剂量组,优甲乐组。除正常对照组外,其余各组每日给大鼠灌服丙基硫氧嘧啶(20 mg·kg-1)连续造模,理气消瘿片低、中、高剂量组同时分别灌服2.4,4.8,9.6 g·kg-1的理气消瘿片水溶液,阳性药物组同时灌以7.8μg·kg-1的优甲乐。60 d后处死动物,留取血清和甲状腺组织。计算大鼠甲状腺指数,检测血清游离三碘甲状腺氨酸原氨酸(FT3),游离甲状腺素(FT4),促甲状腺素(TSH)含量,观察甲状腺组织病理改变,利用免疫荧光法观察甲状腺组织中碱性成纤维细胞生长因子(bFGF),抗凋亡基因B细胞淋巴瘤/白血病2(Bcl-2),转化生长因子β(TGF-β)的表达改变。结果:与正常组比较,模型组大鼠血清中FT3,FT4含量升高(P<0.05),TSH含量降低(P<0.05),甲状腺明显肿大(P<0.01),bFGF,Bcl-2表达明显升高,TGF-β表达明显降低;与模型组相比,理气消瘿片能明显降低甲状腺肿大鼠的甲状腺指数(P<0.05),但对血清FT3,FT4,TSH含量影响不明显,使甲状腺组织病理改变减轻,降低bFGF,Bcl-2表达(P<0.01),升高TGF-β表达(P<0.01)。结论:理气消瘿片具有抗甲状腺肿的作用,其机制与调节bFGF,Bcl-2,TGF-β的表达有关。
Objective: To investigate the mechanism of Qiqi Xiaoying tablet on goiter in experimental rats. Methods: The white closed rats (SD) were randomly divided into 6 groups: normal control group, model group, low, medium and high dose of Liqi Xiaoyan Tablet and Euthyrox group. In addition to the normal control group, the rats in other groups were fed with propylthiouracil (20 mg · kg -1) daily for continuous modeling. The rats in low, medium and high doses of Liqi Xiaoyao tablets were dosed with 2.4, 4.8, 9.6 g · kg-1 of qi Xiaoban aqueous solution, while the positive drug group with 7.8 g · kg-1 of Euthyrox. Animals were sacrificed 60 days later, serum and thyroid tissue were collected. Thyroid index of rats was calculated. Free triiodothyronine (FT3), free thyroxine (FT4) and thyroid stimulating hormone (TSH) were measured in thyroid tissue. The pathological changes of thyroid tissue were observed. Thyroid tissue was observed by immunofluorescence Basic fibroblast growth factor (bFGF), antiapoptotic B cell lymphoma / leukemia 2 (Bcl-2), and transforming growth factor-beta (TGF-beta) Results: Compared with normal group, FT3 and FT4 contents in serum increased (P <0.05), TSH content decreased (P <0.05), thyroid gland enlargement (P <0.01), bFGF and Bcl- (P <0.05), but had no significant effect on the content of FT3, FT4 and TSH in the model group. Compared with the model group, Liqi Xiaopian tablet could significantly reduce the thyroid index (P <0.05) The pathological changes of thyroid tissue were alleviated, the expression of bFGF and Bcl-2 decreased (P <0.01) and the expression of TGF-β increased (P <0.01). Conclusion: Qiqi Xiaopi tablet has the effect of anti-goiter and its mechanism is related to the regulation of the expression of bFGF, Bcl-2 and TGF-β.
