目的:探讨绞股蓝总皂甙对淋巴细胞转化和NDA多聚酶Ⅱ活性的影响. 方法:采用C_(67)BL/6J和BALB/c小鼠淋巴细胞混合培养及淋巴细胞转化实验,按[~3H]TdR和TTP掺入法检测DNA合成和DNA多聚酶Ⅱ活性. 结果:小剂量Gyp(2.5—20 mg L~(-1))促进淋巴细胞转化,并提高DNA多聚酶Ⅱ活性,促进淋巴细胞DNA合成,而大剂量Gyp(>40 mgL~(-1))则呈相反作用. 结论:Gyp能通过调节DNA多聚酶Ⅱ活性调控淋巴细胞转化.
Objective: To investigate the effect of gypenosides on lymphocyte transformation and activity of NDA polymerase II. Methods: Using C_(67)BL/6J and BALB/c mouse lymphocyte culture and lymphocyte transformation experiments, according to [~3H]TdR DNA and DNA polymerase II activity were measured by TTP incorporation. Results: Small doses of Gyp (2.5-20 mg L-1) promote lymphocyte transformation, increase DNA polymerase II activity, and promote lymphocyte DNA synthesis. Large doses of Gyp (>40 mgL-1) showed opposite effects. Conclusion: Gyp can regulate lymphocyte transformation by regulating DNA polymerase II activity.
