目的观察研究VEGF单克隆抗体对动物模型实体肿瘤生长的影响,为临床应用提供实验依据。方法选用BALB/c裸鼠10只,将细胞浓度为5×107个/ml的人舌癌细胞,分别用皮试针接种于裸鼠右前肢背部皮下,0.2ml/只,细胞数量为1×107个,注射完毕后随机分实验组和对照组,每组5只。对照组注射200μl Nacl,实验组注射200μl VEGF单克隆抗体,每天1次,持续28天,处死,进行大体观察和临床试验。结果实验组(VEGF单克隆处理组)移植瘤体积显著低于对照组(P<0.01);肿瘤最终重量低于对照组(P<0.05);实验后期,对照组的体重要明显高于实验组(P<0.05)。结论 VEGF单克隆抗体可显著抑制体内移植瘤的生长。
Objective To observe the effect of VEGF monoclonal antibody on the growth of solid tumor in animal model and provide experimental evidence for clinical application. Methods Totally 10 BALB / c nude mice were used to inoculate human tongue cancer cells with a concentration of 5 × 107 cells / ml. The cells were inoculated subcutaneously with 0.2 ml / 107 were randomly divided into experimental group and control group after injection, with 5 in each group. The control group was injected with 200μl Nacl. The experimental group was injected with 200μl VEGF monoclonal antibody once daily for 28 days and sacrificed for general observation and clinical trial. Results The tumor volume in the experimental group (VEGF monoclonal group) was significantly lower than that in the control group (P <0.01). The final weight of the tumor was lower than that of the control group (P <0.05). At the end of the experiment, the body weight of the control group was significantly higher than that of the experimental group (P <0.05). Conclusion VEGF monoclonal antibody can significantly inhibit the growth of xenografts in vivo.
