目的探讨丁苯酞(NBP)对大鼠脑缺血再灌注损伤脑组织中半胱氨酸天冬氨酸特异性蛋白酶(Caspase)表达的影响。方法用线栓法制作大鼠大脑中动脉脑缺血再灌注模型,大鼠随机按体重分为假手术组、模型组和治疗组。治疗组在大脑中动脉栓塞后15 min内,灌胃丁苯酞80 mg·kg-1。用免疫组织化学法和RT-PCR法检测各组大鼠缺血侧脑组织Caspase-3和Caspase-9的表达。结果与模型组比较,治疗组Caspase-3(0.36±0.05 vs 0.47±0.06)和Caspase-9(0.41±0.06 vs 0.64±0.84)的表达显著下调,差异均有统计学意义(P<0.05)。结论丁苯酞对大鼠脑缺血再灌注损伤后的神经细胞凋亡有抑制作用,其机制可能与抑制Caspase-3和Caspase-9的表达有关。
Objective To investigate the effect of butylphthalide (NBP) on caspase expression in brain tissue after cerebral ischemia-reperfusion injury in rats. Methods Rat middle cerebral artery occlusion (MCAO) model was established by thread occlusion. Rats were randomly divided into sham operation group, model group and treatment group by body weight. The treatment group was given butylphthalide 80 mg · kg-1 within 15 min after the middle cerebral artery occlusion. Immunohistochemistry and RT-PCR were used to detect the expression of Caspase-3 and Caspase-9 in the ischemic brain of each group. Results Compared with the model group, the expressions of Caspase-3 (0.36 ± 0.05 vs 0.47 ± 0.06) and Caspase-9 (0.41 ± 0.06 vs 0.64 ± 0.84) in the treatment group were significantly decreased (P <0.05). Conclusion Butylphthalide can inhibit neuronal apoptosis after cerebral ischemia-reperfusion injury in rats, which may be related to the inhibition of the expression of Caspase-3 and Caspase-9.
